        <p>Остеоартрит является одной из наиболее распространенных причин хронической боли, снижения трудоспособности и инвалидизации, а наследственная предрасположенность существенно влияет на риск поражения крупных суставов. Особый интерес представляют генетические варианты, регулирующие развитие и гомеостаз хрящевой ткани. Цель: проанализировать научные данные о влиянии однонуклеотидных полиморфизмов rs143383 и rs143384 гена фактора дифференцировки и роста 5 (GDF-5) на риск развития, клинические проявления и тяжесть  остеоартрита. Был произведен поиск научных статей за 20 лет в базах данных PubMed/MEDLINE на английском языке и Google Scholar на русском и английском языках по запросам «osteoarthritis GDF5», «rs143383 osteoarthritis», «rs143384 osteoarthritis», «GDF-5 остеоартрит», «rs143383 остеоартрит», «rs143384 остеоартрит». К рассмотрению было принято 45 работ, содержащих данные об указанных полиморфизмах, из которых отобрано 37 публикаций, из них 28 для rs143383 и 9 для rs143384. Полученные данные были структурированы с формированием таблиц для комплексной оценки влияния полиморфизмов с учетом этнических аспектов, эффекторных аллелей и моделей наследования. Показано, что аллель T rs143383 связан со снижением транскрипционной активности гена GDF-5 и чаще ассоциирован с остеоартритом коленного сустава, реже - тазобедренного, суставов кисти, в отдельных исследованиях – с рентгенологической тяжестью заболевания. Aллель C рассматривается как защитный; в целом, rs143383 имеет наиболее убедительную доказательную базу. Полиморфизм rs143384 характеризуется большей популяционной вариабельностью, способностью модулировать эффекты других генов, он ассоциирован с клиническими проявлениями остеоартрита, главным образом коленного сустава, его тяжестью, обнаружены генно-средовые взаимодействия с ожирением. GDF-5 является одним из ключевых генов, определяющих рост и развитие опорно-двигательного аппарата. Его полиморфизмы rs143383 и rs143384 являются одними из наиболее изученных генетических маркеров восприимчивости к остеоартриту. Rs143383 обладает наиболее высоким уровнем доказательности как маркер восприимчивости к остеоартриту.</p>

      <abstract xml:lang="en" lang-variant="translation" lang-source="translator">
        <p>Osteoarthritis is one of the most common causes of chronic pain, reduced work capacity, and disability, while hereditary predisposition significantly influences the risk of large-joint involvement. Genetic variants that regulate the development and homeostasis of cartilage tissue are of particular interest. Objective: to analyze scientific data on the influence of the single-nucleotide polymorphisms rs143383 and rs143384 of the growth differentiation factor 5 gene (GDF5) on the risk of development, clinical manifestations, and severity of osteoarthritis. A search for scientific articles over the past 20 years was performed in the PubMed/MEDLINE databases (in English) and Google Scholar (in both Russian and English) using the following search queries: "osteoarthritis GDF5", "rs143383 osteoarthritis", "rs143384 osteoarthritis", "GDF-5 остеоартрит", "rs143383 остеоартрит", and "rs143384 остеоартрит". A total of 45 papers containing data on the aforementioned polymorphisms were considered for review, of which 37 publications were selected, including 28 for rs143383 and 9 for rs143384. The obtained data were structured into tables to provide a comprehensive assessment of the influence of these polymorphisms, taking into account ethnic aspects, effector alleles, and inheritance models. It was shown that the T allele of rs143383 is associated with reduced transcriptional activity of the GDF5 gene and is more frequently associated with knee osteoarthritis, less frequently with osteoarthritis of the hip and hand joints, and, in some studies, with the radiographic severity of the disease. The C allele is considered protective; overall, rs143383 has the most convincing evidence base. The rs143384 polymorphism is characterized by greater population variability and the ability to modulate the effects of other genes. It is associated with clinical manifestations of osteoarthritis, mainly knee osteoarthritis, as well as with disease severity; gene–environment interactions with obesity have also been identified. GDF5 is one of the key genes determining the growth and development of the musculoskeletal system. Its polymorphisms rs143383 and rs143384 are among the most extensively studied genetic markers of susceptibility to osteoarthritis. Rs143383 has the highest level of evidence as a marker of susceptibility to osteoarthritis.</p>
